Dapoxetine (Sildenafil) is a selective inhibitor of serotonin reuptake in the brain, a critical element of the serotonin system. Unlike other selective serotonin reuptake inhibitors (SSRIs), dapoxetine is a short-acting phosphodiesterase type 5 (PDE5) inhibitor. This means it does not cause a serious side effect like headaches or heartburn. However, its efficacy is not as well-established in the treatment of premature ejaculation (PE).
Uses:
Dapoxetine has not been FDA-approved to treat PE alone.
Dapoxetine comes in tablet and extended-release (ER) capsules. The tablets are taken once daily or once in 24 hours. The extended-release capsules are taken every day and are taken with food or milk to prevent stomach upset and increase the time taken to ejaculate. The tablets can be taken with or without food or milk. One tablet is a single dose. However, a dose of 20 mg may be taken twice daily to achieve maximum effectiveness.
Dapoxetine is not generally considered safe for use during pregnancy. Women who are or may become pregnant should not handle the crushed or broken tablet. Women who are or may become pregnant should not handle the extended-release capsules, as they may break or tear the capsules. Additionally, they should not take the tablet with other drugs that may interact with dapoxetine.
In the United States, Dapoxetine is available in the form of a 30-milligram tablet. A 30-milligram tablet contains 30 milligrams of dapoxetine.
In Europe, Dapoxetine is available in the form of a single 30-milligram tablet. A single 30-milligram tablet contains 30 milligrams of dapoxetine.
The following side effects have been reported with dapoxetine use. Although rare, the following side effects have been reported with dapoxetine: flushing (redness and warmth in the face, neck, and chest), headache, dizziness, abnormal vision, nausea, vomiting, and diarrhea. Some of these side effects may be temporary and resolve on their own.

Drug-Food Interactions:Do not take St. John’s wort (herbal supplement used to treat depression) while taking Dapoxetine. Avoid consuming grapefruit juice within 24 hours of taking Dapoxetine as it could lead to an increase in the level of Dapoxetine in the body.
Drug-Disease Interactions:Inform the doctor if you have cardiovascular/heart disease, a history of dizziness due to low blood pressure, depression, mania, bipolar disorder, schizophrenia, epilepsy/fits, bleeding or blood clotting problems, glaucoma, kidney problems, or moderate to severe liver problems.
